The Horsham trainer's six-year-old was a favourite heading into the race, having won the Werribee Cup in its last run in December last year.
Rupture was caught in a congested front-end, led by Brendon Hearps-trained Vungers, forcing jockey Dean Holland to ride close to the fence.
Approaching the final turn, Holland snuck in of the back of Vungers and broke away, holding out to win the $40,500 race by a length.
Vungers finished second, followed by Mitchell Freedman-trained Fast Plan.
Rupture gifted Preusker his second Warracknabeal Cup win in three years, after stablemate Kapaulenko won the race in 2018.
The result continues Rupture's impressive streak of form since joining Presuker's stable in 2018.
Since returning from a 42-week spell in June 2019, Rupture has placed in eight of its 11 starts, including six wins.
Preusker celebrated another winner on the day, with three-year-old Offtap claiming race two in its first career start.
