Committee member Bruce Hamilton said about 140 riders had signed up for what he described as an extremely challenging tour.
The riders will be aged between 11 and 17 years.
"The tour has been running for about 40 years and we have riders come from all over Victoria, as well as South Australia, Tasmania and New South Wales," he said.
"The Ararat tour has been known as a very tough tour. Usually the stronger climbers usually do better here because of the challenging Carroll's Cutting, which is a steep hill on the way out to Moyston that the riders have to go over."
A king and queen of the hill will be determined on Carroll's Cutting, and Hamilton said there is an interesting story behind the origins of the hill's name which riders could take some solace in as they battle the hill.
"A trophy is provided by Carroll's Saddlery in Ararat because the hill is named after (the owner's) grandfather," Hamilton said.
"He was a contractor and he excavated through the rock, and he nearly went broke because it was one of the hardest rocks in the world apparently. It was so hard that they named it after him."
Road closures will be in place throughout the time trial at 9.30am on Saturday, but local traffic will continue to have access.
The closures will apply to: Moyston Great Western Road from the Ryhmney Road intersection, all the way back to the Moyston township.
"Local people will still be able to get through but we'd prefer it if they could avoid the area, and just be aware that they may be held up," Hamilton said.
Residents may also be waiting longer for their Sunday brunches.
"Normally they get between 130 and 140 riders, and with that there's approximately four or five hundred people coming in for the weekend," Hamilton said.
"Everybody likes to come to Ararat because it's a really challenging course. Now what's happening is a lot of the families are coming up and not only do they stay in the hotels and all of that, but there's a bit of a social network manifesting itself and the Moyston football oval. They camp there Friday and Saturday night."
Murray Culling will be the only representative for Ararat.
