The Rats face off against Dimboola in Ararat this Saturday and coach Taylor Mason said players were ready for a tough encounter.
"We played Nhill last week and we got up on them, but Dimboola beat Nhill by a lot more than we did," she said.
The Rats beat Nhill at the Anzac Day game by 26 goals, but Dimboola defeated Nhill in round one by 51 goals.
"I'm confident with the ability of my team; if we play our best netball we can come out on top," Mason said.
There is added incentive to win the game - it will be star player Megan Shea's 500th game.
"We're definitely all really excited and proud of her," Mason said.
"To play 500 games is huge and we're really proud to have her on our side, so hopefully we can get the win up for her, because I think that would be the way to top it off."
The team will have a full side this round and that win is surely within reach as the Rats' A Grade side is sitting at the top of the ladder.
"We will be at full strength ... we'll have the best ability with anything they try and throw at us," Mason said.
Dimboola is sitting at third, and A Grade player Abbey Greig - who just played her 150th game with the club - said the game's outcome would be a good indicator of the Roos' strength.
"It will be a challenge to see where we are as compared to last year," Greig said.
"It will be good to see how we're faring this year because I feel like we've improved.
"We've got one girl out this time so we won't have our full side."
Greig said she will be treating it like any other game.
"I'll go in with determination and the idea that we're good enough for a win," she said.
She also acknowledged Shea's accomplishment.
"That's an absolutely amazing feat," she said.
