Chalambar are 5-38, chasing 76 for victory against Youth Club after the Clubbers were bowled out for 75 in its first innings.
None of the Chalambar batsmen reached double figures but with five wickets in hand, the team are slight favourites to win going into the second day of play later this week.
In the other game St Andrews sent Tatyoon in to bat first in a decision the team might regret.
Tatyoon posted 135 from its 40 overs with four batsmen reaching double figures to share the load.
Bailey Hosemans fell just short of a 50, dismissed for 48 at the top of the order but the job was done.
Will Hamilton was the only St Andrews bowler to claim multiple wickets, finishing with two.
It makes for an intriguing final day of semi-final action on Friday with all four teams still in contention.
