Police have confirmed the two people who died in yesterday's crash to be a 71-year-old Horsham woman and a 42-year-old Ararat woman.
Initial reports suggested both women were drivers, however, it has since been revealed the Ararat woman was a passenger, and the second car was being driven by a 36-year-old Black Range man.
The male driver was airlifted to the Royal Melbourne Hospital in a critical condition.
The driver of the truck was a 48-year-old Dartmoor man who was arrested and has been assisting police.
EARLIER, 7.15am:
VicRoads have confirmed the Western Highway has reopened after Tuesday's double fatality vehicle accident.
The section of road at Dadswell's Bridge, at the intersection of Golten South Road and the Western Highway, reopened at about 5:30am on Wednesday.
The road is open with speeds reduced to 80km/h due to the collision. Please allow extra time if travelling through the area.
EARLIER, 2.15pm:
VICTORIA police have confirmed two fatalities as a result of the collision on the Western Highway at 12.20pm earlier today.
Detectives from the Major Collision Investigation Unit say a semi-trailer crashed into the rear of a car turning at a rest stop, pushing the vehicle into head-on into another passing vehicle.
The female drivers of both vehicles died at the scene.
A male passenger in one of the vehicles sustained serious injuries and will be airlifted to hospital.
The uninjured male truck driver was arrested and has been assisting police with their inquiries.
The Western Highway will be closed for most of today while detectives examine the crash site.

EARLIER, 1.30pm:
An Ambulance Victoria spokesperson said a male patient at the scene will be airlifted to hospital.
The Ararat Advertiser has received unconfirmed reports of a fatality.
The Ararat Advertiser can not confirm these reports at this time.
There are major traffic delays, with about five kilometres of traffic backed-up heading towards Horsham.
About eight police units are in attendance, three CFA units, SES and Ambulance.

Emergency services are attending the scene of a serious accident on the Western Highway near Dadswell's Bridge.
Calls for assistance were first made at around 12:30pm on Tuesday, with Ambulance Victoria, SES, CFA and Victoria Police all on the scene.
