A Burras win would gift the club its first minor premiership since Minyip and Murtoa settled their rivalry and merged in 1995.
Ararat needs four points to secure third-place, and with it, a double chance come September.
A loss could see the Rats drop to as low as fifth-place.
Burras coach Billie Bibby said her team were entering the match confident.
"We're obviously not going in too cocky, but we are feeling like we're in a pretty good spot considering our win last time," she said.
"We do know that Ararat has a few girls out, but we're not getting ahead of ourselves. It will be a really good game."
Bibby was hopeful the extra pressure wouldn't faze her side.
"The thing that's helped us this year is that we've stayed quite level headed. Throughout the season that has really helped us out a lot," she said.
"If that (minor premiers) is how it ends up that'd be great for the club. I haven't thought about that but if that's something that did happen it would be really good for sure."
There will be plenty of eyes on the result, with both the Horsham Saints and the Warrack Eagles in line for the final place in qualifying final, should the Rats lose.
Ararat coach Taylor Mason said she was treating the game as a "must-win".
"We are looking at the game as a must-win. Obviously, Minyip-Murtoa is a pretty strong side," she said.
"We're staying positive, and we're working hard at training, so hopefully that can pay off."
Megan Shea will miss the game with a calf injury and Mason, herself, remains a doubt with a groin injury.
"All eight of the girls can play three or four positions on the court. Versatility is definitely one of our strengths and when you have players out it's good to have that," Mason said.
The Horsham Saints sit fifth and a win against Dimboola would see them bridge the four-point gap between Ararat.
The Warrack Eagles would also edge past the Rats, should they beat Horsham, and enter into a two-way hit-out that could be decided by the slimmest of margins.
The Saints and the Eagles are level on 44 points and are separated by less than a percentage point.
