The fundraising event will feature a number of classic cars, hot rods and other special vehicles and the convoy will spend the day travelling across the region with plenty of fun ensured.
The cars will gather at Elizabeth Park, Ararat from 10am tomorrow, before departing for Pomonal at about 11am.
After lunch in Pomonal the group will then head to Stawell for a stop at the Picker's Market.
The Cruise for Sophie will aim to raise funds to help local couple Ash Hustler and Paige Mills, whose daughter Sophie Hustler was born on October 9, 2013 at the Royal Women's Hospital with hypo plastic left heart syndrome, small left ventricle and aortic stenosis.
Sophie has endured several heart operations and will require further treatment in the future.
Grampians Grease Monkeys Hotrod Club member Mark Rudolph said tomorrow's event had attracted interest from people as far as Mount Gambier and other towns including Geelong, Horsham, Bendigo and Ballarat.
Mr Rudolph said there could be anywhere from 50 to 100 cars participate and the cruise isn't just restricted to classic cars - anyone can join in.
"We have really been inundated with responses, it has been fantastic," he said.
"If people are prepared to come along and donate some money we're not going to say no to anyone. It is all about having some fun and helping to raise money for a little girl that is in need."
