Investigators have been told a Toyota sedan was travelling north along the Calder Highway about 3.55pm on Friday.
The car left the road and crashed into a tree.
Emergency services attended the scene and the driver of the car, a 74-year-old Charlton woman, was taken to hospital with life-threatening injuries.
The woman died in hospital.
Police urged anyone who witnessed the crash or with further information to call Crime Stoppers on 1800 333 000.
The fatality brings Victoria’s road toll to 171 this year. The road toll was 145 at the same point last year.
