To mark the occasion, an electronic card congratulating preps and their families was sent to schools including Ararat West Primary.
The 31 preps at the school took part in activities based around the number 100 and enjoyed food prepared by each of their parents.
Education Minister James Merlino congratulated the young pupils on the first of what will be many education milestones.
"The first year of school is an exciting time for prep students and their families," he said.
"We're investing almost $4 billion to make Victoria the Education State, so every community has a great local school and every child gets the chance to succeed."
